Previous studies have suggested that depressogenic effects of Alzheimer's disease are independent of its dementing phase and may be attributable to depressive or anxiety disorders preceding the onset of the cognitive impairments. Therefore, the association of a functional polymorphism in the amyloid precursor protein gene (APP), which is linked to Alzheimer's disease (AD), and depressive disorders has been examined. Analysis of APP-16 and APP-swe-variant-17 genotypes was performed by polymerase chain reaction in a sample of 242 AD patients and 175 controls. There was a significant association between APP-16/IVS10nt-5-4 genotype and AD in females, but not in males. There was also an association between the APP-17/ht-7 polymorphism and AD in females, but not in males. A gender effect was observed, but not in the predicted direction, with female AD patients bearing the APP-17/ht-7T allele having significantly fewer years of education than their homozygous APP-17/ht-7C counterparts. The phosphofructokinase (PFK) isolated from the seagrass Zostera muelleri (Linne) Reichenb.f. (eelgrass) has been purified to electrophoretic homogeneity. The purification method used comprised fractional precipitation with ammonium sulphate, heat-treatment of the ammonium sulphate fraction and chromatographic separation on DEAE-cellulose and diethylaminoethyl-Sepharose. The purified preparation had a specific activity of 4.04 micromol ATP/mg protein per minute. The enzyme was a dimer with a molecular weight of 140,000 as determined by gel filtration.
